DefinitionChEBI: An organoammonium salt obtained by reaction of gadoteric acid with one equivalent of 1-deoxy-1-(methylamino)-D-glucitol (meglumine). It is used in magnetic resonance imaging (MRI) in brain (intracranial), spine and associated tissues of paients ages 2 years and older, to detect and visualise areas with disruption of the blood brain barrier and/or abnormal vascularity of the central nervous system.
SynthesisAdd 11.8kg of chloroacetic acid and 12l of purified water to 100L reactor, stir to dissolve, add 15l of water dissolved with 5kg of sodium hydroxide under ice bath, add 3.2kg of rotary cyclotonin suspended in 6l of water to the reaction, warm up to 80~85??, add 6l of water dissolved with 5.2kg of sodium hydroxide dropwise, control the reaction system pH 10~11, react overnight, cool down to room temperature, then add 10.4l of concentrated hydrochloric acid dropwise, adjust the pH to 2~2.5, filter and collect the filter cake and dry to obtain 7.5kg of crude gadoterate glucosamine. After dropwise addition of 10.4l concentrated hydrochloric acid, adjust the pH to 2 ~ 2.5, stirring precipitation, filtration to collect the cake, drying gadoteric acid glucosamine crude 7.5 kg. In a 50L reactor, add gadoteric acid glucosamine crude 7.5 kg, 18 kg of purified water, heating and stirring to dissolve, cooling and precipitation, filtration, washing, drying gadoteric acid glucosamine 6.21 kg, yield 82.7%, which contains the salt amount of 2%. The content of salt was 2%.
