Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs)[1]. 1. Prepare The stock solutions of fluorogenic peptide substrates and stored at -40 °C. 2. Dilute the stock solution∼100-fold into reaction buffers to prepare 2× substrate buffers (400 μM). 3. The enzymes are then incubated for 40 min. 4. The enzymatic activity is subsequently assayed by rapidly mixing 50 μL of the substrate buffer with 50 μL of the enzyme solution in a 96-well clear bottom plates. 5. Record the fluorescence change on a FLEXStation fluorescence plate reader.
