Esomeprazole magnesium trihydrate Usage And Synthesis

Chemical PropertiesOff-White Powder
UsesS-Form of Omeprazole. Gastric proton-pump inhibitor
Usesesomeprazole magnesium trihydrate for the healing of peptic ulcers associated with non-steroidal anti-inflammatory drug NSAID (non-selective and COX-2 selective) therapy.
  1. Zollinger-Ellison syndrome
  2. Peptic ulcer, site unspecified
  3. Duodenal ulcer
  4. Gastric ulcer
  5. Gastro-oesophageal reflux disease
Esomeprazole belongs to the family of medications known as proton pump inhibitors (PPIs). Esomeprazole is registered by the Therapeutic Goods Administration (TGA) for the following indications:
  1. Gastro-Oesophageal Reflux Disease (GORD):
  2. treatment of erosive reflux oesophagitis;
  3. long-term management of patients with healed oesophagitis to prevent relapse;
  4. symptomatic treatment of gastro-oesophageal reflux disease (GORD).
In combination with appropriate antibiotics for:
  1. healing of duodenal ulcer associated with Helicobacter pylori;
  2. eradication of Helicobacter pylori in patients with active or healed peptic ulcer.
  3. Short-term treatment of upper gastrointestinal symptoms associated with non-steroidal anti-inflammatory drug (NSAID) (non-selective and COX-2 selective) therapy.
  4. Healing of gastric ulcers associated with non-steroidal anti-inflammatory drug NSAID (non-selective and COX-2 selective) therapy.
  5. Prevention of gastric and duodenal ulcers associated with non-steroidal anti-inflammatory drug NSAID (non-selective and COX-2 selective) therapy in patients at risk.
UsesEsomeprazole Magnesium Trihydrate is used in preparation of esomeprazole magnesium trihydrate.
Brand nameNexium (AstraZeneca).
Biological ActivityEsomeprazole magnesium trihydrate ((S)-Omeprazole magnesium trihydrate) is a potent, orally active H+, K+-ATPase inhibitor. Esomeprazole magnesium trihydrate has the potential for upper intestinal disorders and gastroesophageal reflux disease research. Esomeprazole magnesium trihydrate acts as an exosome inhibitor by blocking the exosome release by inhibiting V-H+-ATPases.
in vivo

Esomeprazole magnesium (0.5-50 mg/kg; oral gavage; daily; for 10 days; A/J mice) treatment increases gastric total antioxidant capacity and Cu/Zn-superoxide dismutase activity[1].

Animal Model:A/J mice[1]
Dosage:0.5 mg/kg, 5 mg/kg, 50 mg/kg
Administration:Oral gavage; daily; for 10 days
Result:Gastric total antioxidant capacity and Cu/Zn-superoxide dismutase activity are increased.
